Effects of Inhibitors of Ethylene Production on the Activities of Enzymes Related to Ethylene Biosynthesis in Carnation Flowers


Abstract
This study was conducted to investigate the effects of (aminooxy)acetic acid (AOA) and silver thiosulfate (STS) on ethylene biosynthesis and related enzymes¢¥ activities of carnation (Dianthus caryophyllus L.) flowers. ¢¥White Sim¢¥ carnations were harvested at open stage and treated with AOA and STS. The results are summarized as follows:
1. Vase life of STS- or AOA-pretreated flowers was extended more than 2 times as compared with control flowers. This extension of vase life by STS or AOA was derived from the complete inhibition of ethylene production.
2. STS completely inhibited 1-aminocyclopropane-1-carboxylic acid (ACC) accumulation, ACC synthase activity, and ethylene-forming enzyme (EFE) activity of peals ; therefore STS thoroughly inhibited ethylene production of petals. AOA strongly inhibited ACC synthase activity and EFE activity of petals. EFE activity of basal portion of petals was 6 times higher than that of upper portion of petals.
3. ACC content of ovary was little affected by STS or AOA treatment. AOA significantly inhibited EFE activity and ethylene production of ovary, whereas STS partially inhibited EFE activity and ethylene production of ovary.
4. The time-course patterns of ethylene production of petals and ovary were similar to those of EFE activity. Therefore, it is postulated that ethylene production of carnation flowers is controlled mainly by EFE activity.
5. Ethylene production of isolated carnation petals was sustained very low until day 5 and strongly increased at day 6, and then rapidly decreased. STS and AOA completely inhibited ethylene production of isolated petals.
6. EFE activity of isolated carnation petals was sustained very low until day 4 and it reached a peak at day 6, and then rapidly decreased. EFE activity of STS- and AOA-treated petals recorded almost zero.
